  • Cyclosporine, sometimes referred to as "cyclsporin A," is an immunosuppresive agent produced by the soil fungus Beauveria nivia. It initially attracted the interest of the medical community in the management of organ transplant patients, as its mechanism of immune suppression is unique.
  • Cyclosporine is used to prevent rejection of kidney, liver, and heart transplants. It also is used investigationally to prevent rejection problems in bone marrow and other transplants.
  • Cyclosporine suppresses the immune system's T-cells. Both the T-helper cell and the T-suppressor cell may be affected. Cyclosporine also inhibits interleukins.
  • Cyclosporin may be used in combination with another immunosuppressant and a steroid medication.
  • Cyclosporine is generally given once or twice daily depending on the disease being treated.
  • Sandimmune is a soft-gel capsule form of cyclosporine. Absorption from the intestinal tract can vary a lot and is incomplete.
  • Cyclosporine is distributed largely outside the blood volume.
  • The production of a nanosuspension is the transformation of a macrosized powder into a mostly  aqueous nanosized suspension.
  • Cyclosporin A (CsA) is a cyclic oligopeptide that possesses imunomodulating properties and exhibits strong anti-allergic effects. The drug has been used in humans after organ transplantation, and in the last two decades in human dermatology for the treatment of psoriasis and AD.
  • In the last few years, there is a growing interest in the use of CsA to treat canine skin diseases. The efficacy and safety of CsA for the treatment of AD in dogs was evaluated in several trials.
  • CsA at the dose of 5mg/kg was found to have mild adverse reaction, usually GI related, but otherwise was well tolerated.
  • Cyclosporine, used originally to prevent the rejection of transplanted kidneys, continues to be recommended for a variety of organ transplants. However, it has proven effective in treating people with rheumatoid arthritis who have not responded well to other medications as well as those with other rheumatic conditions and severe forms of psoriasis.
  • Renal transplant biopsy is the gold standard for differentiating ailograft rejection from acute tubular necrosis and cyclosponne toxicity.
  • Side effects include high blood pressure, kidney damage, tremors, headaches, seizures, excessive hair growth, excessive gum growth, confusion, coma, and gout.
  • The demand for Cyclosporin in the U.S. market is estimated at $450 million.
